Did you try the unsigned 64bit XP driver posted on their site? Try installing it in compatibility mode, too.

I got it working in Windows 7 64 bit. Here's what I did:
1. Delete and uninstall any drivers you've installed by selecting the device in device manager -- -- Prolific USB-to-Serial Comm Port (should have an exclamation mark on it)
2. Download the PL2303 Prolific DriverInstaller v1.5.0.zip (and extract it somewhere). You can get it here: www.prolific.com.tw/eng/downloads.asp?ID=31
3. Right click on the PL2303_Prolific_DriverInstaller_v1.5.0.exe
4. Choose "Run as Administrator" and follow the install as usual (should work when finished)
