Will I have to reinstall all of my software if I upgrade to Lion?

Short answer - No.

Longer answer - There might be some incompatible software on your Mac which might not work with Lion, but the majority of your programs (and their data) will be copied over during the upgrade.

should be an easy upgrade, but I like to do a clean install on a new HDD (SSD's are getting cheaper and that would help breathe some life into an older Mac)
You can then move the apps you use / data over using a USB enclosure

I do not think so, usually with OSX, you can do an in-place-upgrade. However, based on my experience, "fresh" install always works better than in-place-upgrade. (less problem, better speed, etc.)

I don't think it's possible to upgrade this iMac to Leopard -- is it? Mine has a PowerPC processor, so I don't think it will work. (And if not, is it possible to buy a macmini and use the iMac as a display?) Inquiring minds...
