To me it sounds like the computer is stuck in the headphone mode. You could try plugging in some headphones and gently prodding them while plugged in and the same while taking them out. This might allow the computer to register that the headphones have been unplugged and it would let sound output through the speakers again.

If you can't fix it with gentle prodding pressure then I would be tempted to take it in to the Apple store and see if they will fix it for free.

It simply means the speakers aren't working. The red light is the optical output - it doesn't have anything to do this this problem.

Or, do you mean the speakers work (i.e. the actual speakers on the Mac) when you plug in headphones? Because that would be very weird.

If the above suggestions don't resolve your issue try resetting your PRAM by pressing Command + Option + P + R while rebooting until you hear the signature mac "chime" twice. In the event that doesn't work use a q-tip or toothpick to insert inside the headphone jack in an attempt to switch it back to speaker mode.
