My entire iTunes library lives on my Macbook - what would be the best way to play my iTunes media on my Boxee Box AND stream the media to an Airport Express…
base station in another room simultaneously (since airtunes is not supported)?
Just share your iTunes music folder over your network and add it a location for your media in Boxee.

I use an app called medialink to stream music to video over UPnP to Boxee and other in my house.

The newest Boxee box firmware has AirPlay support. iTunes allows you to play audio simultaneously on multiple devices. You should be able to select both from your MacBook in iTunes.
