I have a friend who also has a first generation iMac with iSight no longer working.
We've tried a number of things, and unfortunately, I think the only recourse is to take it to the Genius Bar and have them look at it (that said, this iMac is now out of Apple Care coverage).
A few things you can try -- reset the SMC or PMU (it's a "simple" process that involves doing some crazy finger yoga). More information on how to do that here: support.apple.com/kb/HT3964
If that doesn't work, you can try to do a few of the trouble shooting options found here:
support.apple.com/kb/ht2090
Unfortunately, if those don't fix your problem -- it's Genius Bar time, and it could cost a pretty penny.
How long has the iSight been broken?
