I think a touchscreen e-Ink device would actually be really hot! The only thing I can see as a major obstacle is the sluggish responsiveness of e-Ink. That said, directly interacting with the content you're reading is a pretty awesome experience (this is something that other touchscreen devices, e.g., iPad, have nailed).
I found find myself touching the screen of my GF's Nook. I simply want to jump from item #1 to item #6 on a list instantly and the screen can keep up with that sort of stuff. Dunno about the feasibility of notes. It already can annotate in line text, but as dave states, the e-ink is too slow to make it feasible.
However, of notes and annotations from the Nook Study app can be transferred over, that could be very useful.
