iPhone OS 4.0 is coming sooner than we thought. Let the speculation begin!
My most wanted features probably won't be much different from anyone else's: some form of multi-tasking, embeddable widgets / live data points that would give a little more glanceable data from the home and unlock screens, and maybe some UI updates to freshen the place up a little. Oh, and one more thing: iPhone OS really, REALLY needs to build trial periods into paid apps.

The new AirMail product from Urban Airship solves that problem in a pretty sweet way in that it gives you an inbox for all of your notifications. urbanairship.com/products/airmail/
I hear you on an updated iTunes. I updated to 9.1 and for some reason it had to "re-optimize" all my 22,000 photos even though I only had 1 folder selected to sync to my iPod.
Better notification. Better battery life. Native GMail push. Multi-tasking, IMO, is going to kill battery life. But it would be nice.
All I ask for is a fixed dictionary (mine keeps changing "me" to "mr", "it's" to "its" etc.), boosted by a customizable dictionary.
Honestly, a UI update is a biggie for me. The only reason I have considered leaving iPhone is because after 3 years, it's kind of boring. The changes don't even have to be anything drastic. (Though, if they rethought and redesigned the springboard, I'm sure no one would mind.)
Of course, multi-tasking is a must.
Beyond those two, and the ones Ryan already listed, I'd love to see some kind of surprise.

Universal inbox (confirmed be Jobs)
Multiple Exchange accounts (for mailboxes only, I see the obvious technical limitation doing it with contacts and calendar)

I have an SMBsettings thingy that allows me to turn it on and off on my 3G. Almost always use it in portrait mode.
It can be locked in software as well. I believe the Kindle app has it.
I just hope that if they add multi-tasking, they also add a way to disable multi-tasking. I'm satisfied with the iPod/Mail/Safari level of multi-tasking as it is currently implemented. If I have to constantly monitor running applications and quit applications that I've left open inadvertently, I'm not going to be very happy with it.

Netflix is on the iPad. As are ebooks of various stripes, as readable as iBooks. But I don't consume as much video on an iPhone as I could, iPads larger size is much better for both netflix and iBooks.
1. Better notification on homescreen (ie: emails, voicemails, txt's, etc.)

I want what I'll never get: Folders for apps. I'm tired of scrolling over 11 pages (or searching) for my apps. I have 20 Spanish apps, 8 tennis apps, 12 fitness apps, 16 games, etc. I want them in folders. For some reason, Jobs believes folders are too complicated for the average user, so they'll never be on iphones or ipads.
And I want multitasking, but I assume that's coming. And I want Flash, but I know hell will freeze over first.

Folders would be really nice. I'm not sure why they haven't been implemented yet. Surely this is a widespread problem. Many people understand the simple folder hierarchy metaphor, maybe they aren't pretty enough.
Some basic multitasking would make both the iPhone and iPad experience so much better. As an example, I'd really like to 'push' tasks from my email into Things. That alone would be awesome.
Sooner than you thought? It's been March the last two times, with a release in June. This is late if anything. Clearly they were just waiting for the iPad to get about.

Multitasking would be great, but what I really want is to be able keep my iPod connected to wi-fi when I lock it without using Insomnia. I'd also like to be able to see the emails I receive in some sort of notification box and I'd definitely like to see an unread mail count somewhere on the status bar.
Oh, I also want percentage on my battery without having to jailbreak. I had it on my iPod before I jailbroke, but after I jailbroke, I had to install a package to show it again.

I think the big ones are:

Considering the laundry list of updates 3.0 had I am somewhat optimistic for version 4.0

What I think would be cool, is if it would almost work like the apple laptops. Where your dock on your iphone works like your laptop, so when you start an app it goes into the dock. It only allows 4-5 apps to work at a time and they are all running, it would help conserve the battery since not all apps can run at a time, but still would allow for safari to load pages while I'm checking my tweet deck and listening to pandora
The only rumor I heard is that it might allow printing. I can see where that's very important for iPad users but I can also see where that would be good for iPhone users in printing email or pulling up a google doc and being able to print it.

Higher resolution support. Printer support. Front cam support. Selected applications can run in the background.
But, I'm still gonna be disappointed since I need file access for the iPhone NOT to feel like a dumbphone. Seriously, I can't download files from the internet? wtf.

lol @ people disliking my comment. Every feature listed/desired in this thread already exists on the android and disliking this comment won't change that fact.
